Indications & Dose: LATENT TUBERCULOSIS PO Adult 300mg/day 6 months Child 5mg/kg OD 6 months | TUBERCULOSIS PO Adult Intermittent therapy: 10mg/kg thrice weekly or 15mg/kg twice weekly, max 900mg/dose | Prophylactic therapy PO Adult 300mg/day 6 months Child 5mg/kg/day, max 300mg/day 6 months | Combination therapy PO Adult Administered during initial & continuation phases of 6 or 8 month regimen: 5mg/kg/day, max 300mg/day or 10mg/kg thrice weekly Child Same as adult dose |
Contra: Hypersensitivity, acute liver disease
Precautions: Renal/hepatic impairment, slow acetylators, epilepsy, history of psychosis, DM, HIV infect, acute porphyria ADR: Serious: Tachycardia, HTN, palpitation, vasculitis, blood disorders, blurred vision, arthralgia, hepatic dysfunction, peripheral neuropathy, hyperbilirubinemia, bilirubinuria, hepatitis, seizures, toxic encephalopathy, Others: Flushing, depression, dizziness, fever, memory impairment, slurred speech, psychosis, rash, gynecomastia, hyperglycemia, metabolic acidosis, pellagra, pyridoxine deficiency, anorexia, epigastric distress, nausea, vomiting, paresthesia, lupus-like syndrome, lymphadenopathy, rheumatic syndrome DDI: Serious Cycloserine adverse CNS effects are increased, Triazolam/Diazepam clearance reduced, Ciprofloxacin increase drug bioavailability, Rifampicin/Methotrexate increases risk of hepatotoxicity, Vincristine increases risk of neurotoxicity, Zalcitabine increases risk of peripheral neuropathy, Alcohol leads to hepatitis, Antidiabetics leads to varies in blood glucose levels, Phenytoin levels increased, Carbamazepine levels rapidly increased & causes toxicity, Prednisolone lowers drug levels, GA metabolism increased, Antacids reduced drug absorption, Paracetamol toxicity raises Diet: 1h before/2h after food Monitor: LFTs, sputum cultures, monthly clinical evaluation, signs of hepatitis |
