Indications & Dose: COLORECTAL CANCER Adjuvant therapy PO Adult 1.25g/m2 BID 2 wk followed by 1 wk rest period, courses of therapy are given in 3 wk cycle for a total of 8 cycles for 6 months | First-line therapy PO Adult 1.25g/m2 BID 2 wk followed by 1 wk rest period, courses of therapy are given in 3 wk cycle | METASTATIC BREAST CANCER Combination therapy PO Adult Initially 2.5g/m2/day in 2 divided doses for 2 wk followed by a 1 wk rest period given in 3 wk cycles. Docetaxel 75mg/m2 over 1h as inf on day 1 of each 3wk cyclepatients with disease who failed to respond to or recurred following anthracycline containing chemotherapy | Mono therapy PO Adult Initially 2.5g/m2/day in 2 divided doses 2 wk, followed by 1 wk rest period, given in 3 wk cyclesresistant to both paclitaxel & chemotherapy regimen that includes anthracycline |
Contra: Hypersensitivity, known deficiency of dihydropyrimidine dehydrogenase, severe renal impairment
Precautions: Hepatic/renal impairment, patients who have received alkylating therapy, elderly, children, patients who have received extensive pelvic radiation, weak/malnourished patients, history of CVD, patients with widespread metastases, patients with depressed bone marrow from radiotherapy/therapy with other antineoplastics ADR: Serious: Palmar-plantar erythrodysesthesia, Edema, blood disorders, bilirubin increased, venous thrombosis, neuropathy, abnormal vision, conjunctivitis, upper GI inflammatory disorders, hemorrhage, viral infect, Others: myalgia, cough, chest pain, headache, lethargy, dizziness, insomnia, mood alteration, depression, nail disorder, rash, skin discoloration, alopecia, erythema, dehydration, motility disorder, oral discomfort, dyspepsia, paresthesia, eye irritation, dyspnea, Fatigue, fever, pain, dermatitis, diarrhea, vomiting, abdominal pain, stomatitis, appetite decreased, anorexia, constipation DDI: Serious Warfarin anticoagulant effects increases markedly, Folic acid enhances drug toxicity, Phenytoin toxicity may occur Diet: With food Monitor: Renal function, CBC with differential & LFTs |
