Indications & Dose: BREAST CANCER Combination therapy/Adjuvant therapy IV Adult 60mg/m2 over 15-20min as inf on days 1 & 8 q28days for 6 cycles with cyclophosphamide & fluorouracil or 100mg/m2 over 15-20min on day 1 q21days for 6 cycles with cyclophosphamide & fluorouracil |
Contra: Hypersensitivity, severe myocardial insufficiency, severe arrhythmias, recent MI, severe hepatic dysfunction, baseline neutrophil count <1500/mm3, previous treatment with anthracyclines up to maximum cumulative doses, patients with CVD
Precautions: Hepatic impairment, previous/recent radiation therapy ADR: Serious: Blood disorders, severe cardiotoxicity, HF, conjunctivitis, Others: nausea, vomiting, diarrhea, mucositis, amenorrhea, alopecia, rash, pruritus, darkening of soles, palms & nails, anorexia, neutropenic fever, hot flashes, lethargy, stomatitis & oesophagitis DDI: Serious Verapamil increases bone marrow depressant toxicity, Ciclosporin increases drug levels, Cimetidine increases exposure to drug, Granisetron potentiates cytotoxic effects of drug, Paclitaxel toxicity raises Monitor: Inj site during infusion, CBC & platelet count, LFTs, SeCr, ECG & LVEF |
