Indications & Dose: TUBERCULOSIS PO Adult 25mg/kg/day, max 2.5g 60 days or until bacteriologic smears & cultures become negative, followed by 15mg/kg/day in patients previously exposed to anti-mycobacterial therapy Child >13 yr: Same as adult dose | Active disease Combination therapy PO Adult Daily therapy: 15-25mg/kg/day, max 1.6g/day, DOT (twice weekly): 50mg/kg, max 4g/dose with pyrazinamide, rifampicin & isoniazid in the initial 2 month phase, followed by a continuation phase of 4 or 7 additional months Child Daily therapy: 15-20mg/kg/day, max 1g/day, DOT (twice weekly): 50mg/kg, max 2.5g/dose with pyrazinamide, rifampicin & isoniazid in the initial 2 month phase, followed by a continuation phase of 4 or 7 additional months |
| Contra: Hypersensitivity, optic neuritis, young children, unconscious patients, patient who unable to discern & report visual changes
Precautions: Renal impairment, ocular disease, elderly ADR: Serious: Thrombocytopenia, hypoglycemia, pulmonary infiltrates, anaphylactoid reactions, TEN, optic neuritis, blurred vision, decreased visual acuity, red-green color blindness, eye pain, eosinophilia, transient hepatic impairment, hyperuricemia, gouty arthritis, bloody sputum, Others: rash, pruritus, fever, joint pain, nausea, vomiting, abdominal pain, GI upset, anorexia, Confusion, disorientation, malaise, dizziness, hallucinations, headache, peripheral neuritis DDI: Serious Antacids reduces absorption of drug Diet: With food Monitor: visual test (>15 mg/kg/day), baseline & periodic renal, hepatic & hematopoietic tests |
