PK: A: Rapid & complete M: Hepatic E: Urine |
Indications & Dose: GOUT PO Adult Initially 250mg BID increased to 500mg BID after 1 wk, if no response, increase by 500mg q4 wk up to 2g/day. Reduce dose by 500mg q6 month if patient is free from acute attacks for at least 6 month |
Contra: Hypersensitivity, high-dose aspirin therapy, blood dyscrasias, uric acid kidney stones, children <2 yrs, penicillin in renal insufficiency patients, renal impairment
Precautions: PUD, salicylates ADR: Serious: flushing, nephrotic syndrome, anaphylaxis, hypersensitivity, exacerbation of acute gouty attack, blood disorders, hepatic necrosis, Others: costovertebral pain, dizziness, fever, headache, alopecia, dermatitis, pruritus, rash, anorexia, nausea, sore gums, vomiting, hematuria, polyuria DDI: Serious Chlorpropamide/Benzodiazepines clearance reduces, Aspirin diminishes drug efficacy, Thiopental increases drug level, Allopurinol increases renal excretion of active metabolite of drug, Fibrates/Diprophylline level increases, Cephalosporins levels increases, Dapsone markedly raises serum drug levels, Nitrates reduces penicillins excretion, Famciclovir/Aciclovir/Ganciclovir/Valaciclovir/Valganciclovir renal excretion reduces & increases plasma levels, Rifampicin/Methotrexate toxicity increases, Zidovudine toxicity occurs Diet: With food Monitor: Uric acid, CBC, renal function test |
