Indications & Dose: HODGKIN’S DISEASE IV Adult Initially 3.7mg/m2, increase to 18.5mg/m2 as single agent (not more often than q7days), if needed MD is given q7days provided white cell counts permit, max dose based on patients tolerability without leucopenia Child Initially 6mg/m2, do not administer more frequently than q7days | LETTERER-SIWE DISEASE IV Child Initially 6.5mg/m2, do not administer more frequently than q7days | MALIGNANT NEOPLASMS IV Adult Initially 3.7mg/m2, increase to 18.5mg/m2 as single agent (not more often than q7days), if needed MD is given q7days provided white cell counts permit, max dose based on patients tolerability without leucopeniaof breast, bladder, kidney, non-small cell lung cancer, choriocarcinoma & kaposi’s sarcoma | TESTICULAR CANCER IV Child Initially 3mg/m2, do not administer more frequently than q7days |
Contra: Hypersensitivity, granulocytopenia, active bacterial infect, IT route of administration, extravasation, elderly patients with cachexia/skin ulcers
Precautions: Hepatic impairment, IHD, pulmonary dysfunction, renal disease with HTN, malignant-cell infiltration of bone marrow, neuromuscular disease ADR: Serious: bone marrow depression, Raynaud’s phenomenon, bronchospasm, tissue necrosis, peripheral/autonomic neuropathy, seizures, hemorrhagic colitis, depression, orthostatic hypotension, paralytic ileus, urinary retention, HTN, Others: myalgia, hyperuricemia, photosensitivity, alopecia, nausea, vomiting, stomatitis, constipation, taste disturbances DDI: Serious Mitomycin acute pulmonary toxicity, Vaccines (Live) diminishes vaccine efficacy, Itraconazole/Erythromycin/Tacrolimus/Voriconazole Increases vinblastine toxicity, MAOIs orthostatic hypotension, Ritonavir severe neutropenia Monitor: CBC, platelet count, BP, pulse, LFT, respiratory status, signs of neuropathy, bone marrow depression, hyperuricemia, anemia, stomatitis |
