Indications & Dose: MALIGNANT PLEURAL MESOTHELIOMA Combination therapy IV Adult 500 mg/m2 on day 1 of each 21 day cycle with cisplatinPre-treatment: Dexamethasone for 3 days 4mg BID PO.; start 1 day before; Folic acid 350-1000µg/day PO; start 1 wk before, continue till 3 wks after]; & Vit B12 1000µg IM start 1 wk before, and then every 3 cycles. New treatment cycles should not begin unl | NON-SMALL CELL LUNG CANCER Combination therapy IV Adult Initially 500 mg/m2 on day 1 of each 21 day cycle with cisplatin.For maintenance or second line treatment 500 mg/m2 on day 1 of each 21 day cycle |
| Contra: Severe hypersensitivity
Precautions: hepatic dysfunction, on nephrotoxic drugs (aminoglycosides, cisplatin, loop diuretics, ciclosporin, and drugs that are secreted by renal tubules, such as probenecid, penicillin) ADR: Serious: ARF, MI, cerebrovascular events, increased ALT, AST, anemia, leukopenia, neutropenia, Others: edema, hepatitis, colitis, intestinal pneumonitis, fatigue, rash, nausea/vomiting, stomatitis DDI: Others NSAIDs/Aspirin in high doses decreases pemetrexed elimination Monitor: CBC with differential & platelets, serum creatinine, BUN, total bilirubin, ALT, AST |
