Indications & Dose: ABETALIPOPROTEINEMIA PO Adult 50-100mg/kg (as dl-alpha tocoferil acetate) or 33-67mg/kg (as d-alpha tocopherol) Child Neonate: 100mg/kg OD, 1 month–18 yr: 50–100mg/kg OD | CYSTIC FIBROSIS PO Adult 100-200mg (as dl-alpha tocoferil acetate) or 67-135mg (as d-alpha tocopherol) | MALABSORPTION IN CYSTIC FIBROSIS Combination therapy PO Child 1 month–1 yr: 50mg OD, 1–12 yrs100mg OD, 12–18 yr: 100-200mg OD adjust as needed (with food and pancreatic enzymes) | VITAMIN E DEFICIENCY PO Adult 60-75U/day Child Neonate: 10mg/kg OD, 1 month–18 yr: 2-10mg/kg/day max 20mg/kg | PO Child In cholestasis & severe liver disease: Neonate: 10mg/kg/day, 1 month-12 yr: Initially 100mg/day, adjust according to response max 200mg/kg/day, 12-18 yr: Initially 200mg/day, adjust according to response max 200mg/kg/day | Prophylactic therapy PO Adult 30U/day | IM Child In cholestasis & severe liver disease: Neonate: 10mg/kg once a month; 1 month-18 yr: 10mg/kg max 100mg once a month |
| ADR: Serious: Blurred vision, increase bleeding tendency in vitamin-K deficient patients, necrotising enterocolitis (premature infants weighing <1.5 kg), Others: Diarrhea (large dose), abdominal pain, GI disturbances, dizziness, fatigue, weakness, contact dermatitis (topical)
DDI: Serious Orlistat decreases vitamin E absorption, Dicoumarol effects slightly increases, Warfarin may causes bleeding |
