Indications & Dose: FEMALE HYPOGONADISM Consolidation therapy PO Adult Conjugated: 0.3-0.625mg/day given in cyclic regimen (3 wk on drug & 1 wk off). Subsequent doses should be adjusted according to severity of the disease and response. Esterified: 2.5-7.5mg/day in divided doses for 20 days followed by 10 drug free days, may be repeated if needed | INOPERABLE METASTATIC BREAST CANCER IN MEN/POSTMENOPAUSAL WOMEN Palliative therapy PO Adult Conjugated/Esterified: 10mg TID 3 month | INOPERABLE PROSTATE CARCINOMA Palliative therapy PO Adult Conjugated/Esterified: 1.25-2.5mg TID | OSTEOPOROSIS Prophylactic therapy PO Adult Conjugated: 0.3mg/day in a continuous daily regimen or in a cyclic regimen (25 days on drug & 5 day off), repeated as required | OVARIECTOMY/PRIMARY OVARIAN FAILURE PO Adult Conjugated/Esterified: 1.25mg/day given in cyclic regimen (3 wk on drug & 1 wk off). Subsequent doses should be adjusted according to severity of the disease & response. | UTERINE BLEEDING DUE TO HORMONAL IMBALANCE Emergency therapy IV/IM Adult Conjugated: 25mg repeated in 6-12h | VASOMOTOR MENOPAUSAL SYMPTOMS Moderate-to-severe PO Adult Conjugated: 0.3mg/day as continuous daily regimen or cyclic regimen (25 days on drug & 5 day off), repeated as required. Synthetic conjugated: Initial dose 0.45-1.25mg/day, adjust the dose based on response. Contra: Undiagnosed abnormal vaginal bleeding, active/recent arterial thromboembolic disease, venous thromboembolic disorders, hepatic dysfunction, estrogen dependent neoplasms, rotor syndromes
Precautions: Antiphospholipid antibodies or predisposing factors to thromboembolism, migraine, history of breast nodules/fibrocystic disease, uterine fibroids, history of endometriosis, history of cholestatic jaundice, asthma ADR: Serious: Endometrial carcinoma, palliation of cancers, cholestatic jaundice, withdrawal bleeding, hypercalcemia, gynecomastia, venous & arterial thrombosis, breast cancer, hypersensitivity reactions, Others: Nausea, vomiting, abdominal cramps, weight changes, dizziness, changes in libido, migraine, breast enlargement & tenderness, depression, mood changes, headache, leg cramps, vaginal candidiasis, decreased tolerance of contact lenses, local irritation, rhinorrhea, epistaxis, fluid retention, glucose intolerance, altered blood lipids, premenstrual-like syndrome, rash DDI: Serious Theophylline clearance is reduced, Phenobarbital/Carbamazepine/Rifampin decreases drug levels leads to decreases therapeutic effects, Ursodeoxycholic Acid/Anticoagulants efficacy decreased, Diltiazem increases drug levels, Erythromycin/Clarithromycin/Itraconazole/Ketoconazole increases drug levels leads to increase incidence of adverse effects, Corticosteroids metabolism decreased, Imipramine toxicity raises |
