Indications & Dose: MANTLE CELL LYMPHOMA IV Adult 1.3 mg/m2 twice weekly for 2 wk (days 1, 4, 8 & 11), followed by 10-day rest period (days 12 to 21). Allow at least 72h to elapse between doses. One treatment cycle equals 21 days. For extended therapy of more than 8 cycles, drug may be given either on the standard schedule recommended above or on a maintenance schedule of one dose weekly for 4 wk (days 1, 8, 15, and 22) followed by 13 day rest period (23-35) | RELAPSED MULTIPLE MYELOMA IV Adult 1.3 mg/m2 twice weekly for 2 wk (days 1, 4, 8 & 11), followed by 10-day rest period (days 12 to 21). Contra: Hypersensitivity, acute diffuse infiltrative pulmonary disease, pericardial disease
Precautions: DM, CVD, pulmonary disease, history of seizures, amyloidosis, risk of neuropathy, hepatic/renal impairment, children ADR: Serious: Edema, cardiac disorder, hypotension, psychiatric disturbance, peripheral neuropathy, respiratory tract infect, Herpesvirus infect, HF, blood disorders, hypercalcemia, bleeding events, Others: Inj site irritation, weakness, anxiety, dehydration, paresthesia, arthralgia, limb pain, bone pain, myalgia, muscle cramps, rigors, dyspnea, cough, dysesthesia, headache, insomnia, dizziness, rash, nausea, diarrhea, constipation, anorexia, vomiting, abdominal pain, abnormal taste DDI: Serious Antihypertensives causes additive hypotension, Thalidomide causes peripheral neuropathy, Ketoconazole increases drug levels, Ciclosporin increases the risk of severe neuropathy, Antidiabetics leads to hyper-/hypoglycemia Monitor: Signs/symptoms of peripheral neuropathy, dehydration, hypotension, reversible posterior leukoencephalopathy syndrome, CBC with differential & platelets, renal function, pulmonary function, LFTs |
