Indications & Dose: LOIASIS PO Adult 1mg/kg as single dose on day 1, doubled on two successive days, then adjusted to 2–3 mg/kg TID 18 days | Prophylactic therapy PO Adult 300mg weekly for as long as exposure occurs | LYMPHATIC FILARIASIS (BANCROFTIAN) PO Adult 1mg/kg single dose on day 1, increase gradually over 3 days to 6mg/kg/day 12 days Child <10 yr: Half the adult dose, >10 yr: Same as adult dose | LYMPHATIC FILARIASIS (BRUGIAN) PO Adult 1mg/kg single dose day 1, increased gradually over 3 days to 3–6mg/kg/day in divided doses 6–12 days Child <10 yr: Half the adult dose, >10 yr: Same as adult dose | OCCULT FILARIASIS PO Adult 8mg/kg/day 14 days, repeat as needed if symptoms return |
Precautions: Onchocerciasis, renal impairment, cardiac disorders, severe acute diseases
ADR: Serious: hypersensitivity, hematuria, Others: urticaria, nausea, vomiting, headache, dizziness, drowsiness, fever, joint pain, anorexia, malaise DDI: Serious Ammonium chloride increases drug loss in urine, Sodium Bicarbonate markedly increases drug retention Diet: After food |
