Indications & Dose: ACNE TP Adult Gel (5%): Applied to the affected area BID | DERMATITIS HERPETIFORMIS PO Adult 50mg/day, gradually increased to 300mg/day or more if required | MULTIBACILLARY LEPROSY PO Adult 50-70kg: Dapsone 100 mg/day along with rifampicin (600mg/month), clofazimine (300mg/month & 50mg/day) for 1 yr, <35 kg: Decrease rifampicin & dapsone dose (50mg or 1-2mg/kg/day) Child 10-14 yr: Dapsone 50mg/day, rifampicin (450mg/month ), clofazimine (150mg/month & 50mg q2days for 1 yr | PAUCIBACILLARY LEPROSY PO Adult Dapsone 100mg/day with rifampicin 600mg once a month for 6 month Child 10-14 yr: Dapsone 100mg/day with rifampicin 450mg once monthly |
Contra: Hypersensitivity
Precautions: Severe anemia, G6PD deficiency, hemoglobin M deficiency, methemoglobin reductase deficiency ADR: Serious: Tachycardia, SJS, TEN, interstitial pneumonitis, pulmonary eosinophilia, nephrotic syndrome, renal papillary necrosis, tinnitus, blurred vision, cholestatic jaundice, hepatitis, peripheral neuropathy, hypoalbuminemia, blood disorders, Others: fever, headache, insomnia, psychosis, vertigo, abdominal pain, nausea, vomiting, bullous & exfoliative dermatitis, erythema nodosum, phototoxicity, urticaria, infectious mononucleosis-like syndrome DDI: Serious Clofazimine anti-inflammatory effects is reduced, Probenecid/Cimetidine increases drug levels, Rifampicin/Rifabutin increases drug toxicity & its clearance, Zidovudine increases risk of blood dyscrasias, Trimethoprim leads to increases both levels, efficacy & drug toxicity raises Diet: After food Monitor: G6PD levels prior to initiation, signs of jaundice & hemolysis, CBC |
