Indications & Dose: JUVENILE IDIOPATHIC ARTHRITIS PO Child 1 month-18 yrs: 5-6.5mg/kg/day, max 400mg/day | LUPUS ERYTHEMATOSUS/LIGHT-SENSITIVE SKIN ERUPTIONS PO Adult Initially 400mg BID, MD 200-400mg/day, max 400mg/day. Discontinue the drug if there is no improvement after 6 month Child 6.5mg/kg/day or 400mg/day | MALARIA Prophylactic therapy PO Adult 400mg q7days Child 6.5mg/kg, max 400mg | Acute PO Adult 800mg single dose or initially 800mg followed by 400mg after 6-8h & further 400mg on each of the 2 following days Child Initially 13mg/kg followed by 6.5mg/kg after 6h & again on 2nd & 3rd days | RHEUMATOID ARTHRITIS PO Adult Initially 400-600mg/day, MD 200-400mg/day, max 400mg/day. Discontinue the drug if there is no improvement after 6 months Child 6.5mg/kg/day or 400mg/day |
| Contra: Hypersensitivity, retinal/visual field changes attributable to 4-aminoquinolines, long-term use in children
Precautions: Known G6PD deficiency, hepatic/renal impairment, porphyria, psoriasis, myasthenia gravis, GI disorders ADR: Serious: seizure, angioedema, exfoliative dermatitis, SJS, tinnitus, bronchospasm, abnormal color vision, abnormal retinal pigmentation, nystagmus, blood disorders, Others: myopathy, corneal changes/deposits, urticaria, nausea, vomiting, diarrhea, pigmentation changes, rash, alopecia, ataxia, dizziness, emotional changes, headache, irritability, lassitude DDI: Serious Antacids reduces absorption of drug, Cimetidine reduces metabolism & clearance of drug Diet: With food Monitor: Ophthalmologic exam, CBC at baseline & periodically, muscle strength during long-term therapy |
