Indications & Dose: CONGENITAL SYPHILIS IM Child Neonates >1.2 kg: 50, 000U/kg as single doseAsymptomatic | GROUP A STREPTOCOCCAL UPPER RESPIRATORY INFECTION IM Adult 1.2MU single dose Child Primary prevention of rheumatic fever: <27 kg: 600, 000U single dose, >27 kg: 1.2MU single dose, Secondary prevention of rheumatic fever: <27 kg: 600, 000U q3-4 wk, >27 kg: 1.2MU q3-4 wk | SECONDARY PREVENTION OF GLOMERULONEPHRITIS IM Adult 1.2MU q4 wk or 600, 000U twice monthly | SECONDARY PREVENTION OF RHEUMATIC FEVER IM Adult 1.2MU q3-4 wk or 600, 000U twice monthly | SYPHILIS IM Adult 2.4MU single dose in 2 injection sites for primary, secondary, early latent (<1 year duration) Child Infants and Children: 50, 000U/kg single Inj, max 2.4MU for primary, secondary, early latent (<1 year duration) | IM Adult 2.4MU in 2 injection sites once weekly 3 doses for late latent, latent with unknown duration Child 50, 000U/kg every wk 3 doses, max 2.4MU/dose Late latent, latent with unknown duration |

Precautions: Asthma, renal impairment, history of seizure ADR: Serious: anaphylactoid/hypersensitivity reactions, cardiac arrest, CVA, hypotension, seizure, severe agitation, tinnitus, intestinal necrosis, arthritis exacerbation, blindness/blurred vision, renal impairment, diaphoresis, Others: nausea, vomiting, impotence, anxiety, confusion, dizziness, headache DDI: Serious Methotrexate causes acute toxicity, Indomethacin/Aspirin/Phenylbutazone/Sulfinpyrazone prolongs drug half-life, Tetracyclines reduces penicillins efficacy, Probenecid reduces penicillins excretion Monitor: Anaphylaxis during first dose |
